Working Principle: There are two separate light paths: bottom (Transmitted light) and top (Reflected light) illumination. The bottom illumination system is preferred for examining transparent paintable materials that can transmit more light. It is mostly used to visualize biological samples by staining them with various histochemical or immunohistochemical dyes. In the top illumination system, it is used to visualize opaque samples that do not transmit light. Here, the light hits the sample and reflects back to the objective according to the color and brightness characteristics of the sample surface, and an image is formed on the microscope. This system is mostly preferred for samples in the material field.
